Both Sanchez and Matic missed the first leg at Old Trafford – which Barca won 1-0 – but were pictured travelling with the United squad to Spain on Monday.
Sanchez has not played for United since the start of March, when he suffered ligament damage in the Premier League victory over Southampton.
Matic, meanwhile, has been absent for the past two weeks.
Speaking after arriving in Barcelona, Solskjaer said: ‘Fitness wise they are both available and of course for Alexis, who has played for this club before, it’s a big night.
‘When he’s on the pitch I’m sure he will want to prove a point. Not just to Barcelona but also to us as well because he’s been injured and he’s not had a great season.
‘Hopefully he can finish the season well because this is the business end. Let’s hope Sanchez can be happy with his performance.’
On United’s hopes of progressing in the Champions League, Solskjaer added: ‘Well of course the memory of PSG will help the players.
‘As a team we know that we can turn things around. Barcelona is difficult prospect but PSG is not that long ago and we are one goal better off this time around.’
Diogo Dalot, who started last week’s first leg, is also optimistic about United’s chances of reversing their 1-0 deficit at the Nou Camp.
‘I think we proved that we’re a team who never gives up,’ he said.
‘After losing the first leg we’ll come here to see if we can level the game and then try to win it – which is what’s in our minds at all times.’
